WHY

I have paladin and I wanted to amp my weapon So first I bought sign of imperishability but when I try to use it the "USE" button is not active, So whys that???

You must use spheres as well as signs. For example, if I wanted to amp my lv13 light boilersuit using a sign I would first need to acquire a [sphere of Defence Enhancement II] as well as a [sign of Imperishability].. Then select sphere from inventory and click "use" then select which piece of armor, then the next prompt will give you three options for "Item spoiling insurance". This is where you may select "Use sign of Imperishability", then you can click "Amplify".

Now am using the sign while am amping but it always says nothing happened !!! why is that???

You dont have enough money, each amplification has a cost in gold too (you can read it in the lower part of the amplification menu).
